Negative Staining

TEM Protocol- Negative Staining

  1. Need a grid that has formvar and carbon coatings. Ted Pella product number 01801 works very well and is ready to use. 01801 is a copper grid of 200 mesh.
  2. If the grid package has been opened for a while I recommend that the grids be exposed to U.V. light for no more than 5 minutes.
  3. Place 10 µl of sample on grid for 2 minutes. Use whatman paper  to wick off, be careful to be gentle and only touch the side of the grid.
  4. Do 2 washes of autoclaved MegPure water. Again use whatman paper to wick off washes.
  5. Place 10 µ1 1% Uranyl Acetate on grid for 2 minutes. 0.05g of Uranyl Acetate added to 10ml of autoclaved MegPure water.
  6. Wick off excess stain with whatman paper.
  7. Air dry.
Note: Depending on your sample, a solution of 2 - 0.5 % Uranyl Acetate can be tried.
